Can be done in either but most pro illustrators prefer Illustrator because of, aside from the catchy name, its vector capabilities.
Illustrator creates vector graphics and vector text. Vectors consist of lines and curves that contain mathematical objects that can easily be scaled by computer to any size without losing quality. This is a preferred format for print media, which illustrations are usually meant for; ie. posters, t-shirt and magazine print.
Photoshop creates bitmap images based on pixels.
Bitmapped graphics are measured by the number of dots per unit - typically referred to as dpi (dots per inch). These bitmap graphics are much larger in file size than vector graphics and are subject to diminished change in quality if you enlarge it or reduce it. Bitmap images are superior in terms of blends of color, spectrum and definition.
